Are single letters a good investment? not just in Hindi but in a variety of Indian languages like Tamil, Bengali, etc? What are people's opinions?

I have all of the above.

There tend to be a lot more "letters" than in English. This is because similar to Hirigana, each character combines a constanant with a vowel.

You can get an idea of which are the most commonly used ones by doing a simple Google search.

I have to say I am surprised that there might be any left.
